The histochemical investigation on the periodontal ligament in excessive cortisone administered rabbit


Abstract
The purpose of this investigation was to observe the effect of the excessive administration cortisone on the fibrous compoments of periodontal membrane.
The forty experimental animals weighing approximately 1 Kg were divided into a group of eight normal controlls and a group of thirty two cortison administered groups.. The cortisone administered groups. The cortisone administered animal groups were given daily subcutanious injection of 10 mg, 15mg, 20mg, 25mg, for 7, 14, 21 and 28days. The experimental animals were sacrificed under anesthesia. The speciments were removed from molar region of mandible and were fixed in the 10% formalin and Carnoy¢¥s solution. Decalcification of the specimente were made with 3% formic acid and they were embedded in celloidin.
The histological and histochemical procedure employed were Van Gieson stain, Bielschowsky -Gomori¢¥s silver impregnation method, Masson-Gomori¢¥s trichrom stain, Malloy¢¥s azan stain, alloxanShiff stain, PAS reaction and hematoxylin eosin stain.
The results were as follows:
1) For the animals received 25 mg/Kg of cortisone per day for from 14 to 2ldays, the arrangement
of Sharpey¢¥s fibers were irregular in the upper portion of alveolar crest.
2) The argyrophil fibers of administered animals were weakened after being injected of cortisone
20mg/Kg for 28 days
3) The stain ability of fibrous elements in The area with high affinity to hemat lin in the
periphery of the pericemental space were decreased for the hematoxlin as the amount of cortisone injection increased for the PAS and alloxan-Schiff reaction.
4) The more the cortisone was given, the thicker the periocemertal space
